is it possibile to set a playlist of local mp4 files to play with mediamanager?
is it possibile to play a random file at a time from a local directory of mp4 files?

The Media Manager inserts mp4 files using the HTML <video> tag, and playlists are not a feature of the <video> tag.

You could conceivably create a playlist using javascript, but this would fulfill your requirement to play files from a local folder.

There may a dedicated Joomla extension that can provide a <video> playlist - https://extensions.joomla.org/category/multimedia/multimedia-players/
